2. PRODUCT DESCRIPTION
Paratru-250 Suspension is a paediatric-grade Paracetamol Oral Suspension I.P., delivering 250 mg of Paracetamol per 5 ml in a palatable flavoured syrup base. It is indicated for the symptomatic relief of fever (pyrexia) and mild to moderate pain in infants, children, and adolescents. As one of the safest and most widely used analgesic-antipyretic agents in paediatric medicine, Paracetamol remains the WHO first-line recommendation for children’s fever management globally.
Paratru-250 Suspension is formulated for ease of administration in paediatric patients who cannot swallow solid dosage forms. The pleasant flavour and ready-to-use suspension format ensure high compliance in children. 3. MECHANISM OF ACTION
Antipyretic Action
Paracetamol reduces fever by acting on the hypothalamic heat-regulating centre. It inhibits prostaglandin synthesis (particularly PGE2) in the central nervous system by blocking cyclooxygenase (COX) enzyme activity within the CNS. This lowers the hypothalamic set-point temperature, causing peripheral vasodilation and increased heat dissipation — resulting in measurable reduction of body temperature within 30–60 minutes of oral administration.
Analgesic Action
Paracetamol exerts its analgesic effect through a combination of central and peripheral mechanisms. Centrally, it inhibits spinal cord prostaglandin synthesis, reducing pain signal amplification. It also activates descending serotonergic inhibitory pathways, modulating pain perception. Additionally, recent research suggests Paracetamol may interact with the endocannabinoid system via its active metabolite AM404, contributing to its analgesic properties.
Safety Profile
Unlike NSAIDs (Ibuprofen, Naproxen), Paracetamol does not inhibit peripheral COX enzymes significantly at therapeutic doses, making it safe for gastric mucosa, platelets, and renal function. It does not carry the risk of Reye’s syndrome (unlike Aspirin), making it the preferred choice in paediatric fever and pain management.
